Validasi Udang Air Tawar <i>Caridina gracilipes</i> Danau Sentani Menggunakan Pendekatan DNA Barcoding

Danau Sentani merupakan danau terbesar di Provinsi Papua yang masuk dalam wilayah pemerintahan Kota Jayapura dan Kabupaten Jayapura. Salah satu sumberdaya perikanan penting yang belum dimanfaatkan dengan baik di daerah Danau Sentani adalah udang air tawar. Informasi kejelasan dari suatu spesies, terutama sumberdaya perairan Danau Sentani, khususnya udang air tawar Caridina gracilipes masih sangat minim. Oleh karena itu penelitian ini dilakukan dengan tujuan untuk melakukan validasi secara genetik udang air tawar Caridina gracilipes yang berada pada perairan Danau Sentani. Metode yang digunakan adalah metode Barcoding DNA Mitokondria, gen COI. Gen COI ketiga sampel berhasil diamplifikasi dengan panjang basa 767 (SpS01), dan 686 (SpS02 dan SpS03). Hasil pensejajaran pada Gen Bank NCBI dengan aplikasi BLAST menunjukkan kemiripan sampel nukleotida SpS01 99.54%, SpS02 sebesar 99.85% dan SpS03 sebesar 100% dengan data GenBank NCBI yaitu Caridina gracilipes. Hasil BLAST diperkuat juga dengan pohon filogenetik, yang menunjukkan bahwa ketiga sampel (SpS01, SpS02, dan SpS03) berada pada satu kelompok spesies Cardina gracilipes.
